mRNA diffusion explains protein gradients in \textit{Drosophila} early development

چکیده
We propose a new model describing the production and the establishment of the stable gradient of the Bicoid protein along the antero-posterior axis of the embryo of Drosophila. In this model, we consider that bicoid mRNA diffuses along the antero-posterior axis of the embryo and the protein is produced in the ribosomes localized near the syncytial nuclei. Bicoid protein stays localized near the syncytial nuclei as observed in experiments. We calibrate the parameters of the mathematical model with experimental data taken during the cleavage stages 11 to 14 of the developing embryo of Drosophila. We obtain good agreement between the experimental and the model gradients, with relative errors in the range 5 − 8%. The inferred diffusion coefficient of bicoid mRNA is in the range 4.6 × 10−12−1.5×10−11 m2s−1, in agreement with the theoretical predictions and experimental measurements for the diffusion of macromolecules in the cytoplasm. We show that the model based on the mRNA diffusion hypothesis is consistent with the known observational data, supporting the recent experimental findings of the gradient of bicoid mRNA in Drosophila [Spirov et al. (2009) Development 136:605-614].

متن کاملIntegrin Linked Kinase (X-ILK) Function during Embryonic Development and within Adult Tissues of Xenopus laevis
Integrin linked kinase (ILK) is a serine/threonine protein kinase implicated in the phosphatidylinositol 3’kinase (PI3’K) pathway. Integrin linked kinase has been investigated in different organisms such as mammalian systems (human, mice, rat), insects (Drosophila) and nematodes (Cenorhabditis elegans), however to date little data regarding ILK research on amphibians has been reported. In...
متن کاملTime Course of Degradation and Deadenylation of Maternal c-mos and Cyclin A2 mRNA during Early Development of One-Cell Embryo in Mouse
Early in the development of many animals, before transcription begins, any change in the pattern of protein synthesis is attributed to a change in the translational activity or stability of mRNA in the egg and early embryo. As a result, translational control is critical for a variety of developmental decisions, including oocyte maturation and initiation of preimplantation development. In this s...
متن کاملStochastic Spatio-Temporal Dynamic Model for Gene/Protein Interaction Network in Early Drosophila Development
In order to investigate the possible mechanisms for eve stripe formation of Drosophila embryo, a spatio-temporal gene/protein interaction network model is proposed to mimic dynamic behaviors of protein synthesis, protein decay, mRNA decay, protein diffusion, transcription regulations and autoregulation to analyze the interplay of genes and proteins at different compartments in early embryogenes...
متن کاملKnown maternal gradients are not sufficient for the establishment of gap domains in Drosophila melanogaster
Gap genes are among the first transcriptional targets of maternal morphogen gradients in the early Drosophila embryo. However, it remains unclear whether these gradients are indeed sufficient to establish the boundaries of localized gap gene expression patterns. In this study, we address this question using gap gene circuits, which are data-driven mathematical tools for extracting regulatory in...
